Enhancement of intranasal mucosal immunization of mucosal vaccines by ultrasonic treatment

The pathogens of most infectious diseases invade the host through mucosal sites, and immunization with vaccines is best means combating these diseases. Oral delivery nasal are common methods vaccination. However, process inefficient, vaccination ineffective because vaccine formulation easily rapidly removed has difficulty in crossing surface. In this paper, we investigated whether immune response could be enhanced by ultrasound facilitation preparations. For purpose, used manganese dioxide (MnO2) as carrier/adjuvant, coated chitosan oligosaccharide (COS) to enhance adsorption, further physically adsorbed model antigen ovalbumin (OVA) construct a nanoparticulate MnO2@COS@OVA. Ultrasound treatment was found promote recruitment dendritic cells (DCs) macrophages well T-cell infiltration tissues studies. With assistance, MnO2@COS@OVA particles promoted maturation DCs vitro vivo production effector memory T cytokine secretion splenocytes vitro. particular, significantly increased levels secretory IgA antibodies mucosa genital tract experimental mice. addition, data showed that had good biocompatibility caused no significant damage vital organ tissue. These suggest can induction efficient responses provide new ideas for opening clinical translation vaccines.
